ScientificBackground | 8-hydroxy-2-deoxyGuanosine(8-OH-dG)isproducedbytheoxidativedamageofDNAbyreactiveoxygenandnitrogenspeciesandservesasanestablishedMarkerofoxidativestress(1-4).Hydroxylationofguanosineoccursinresponsetobothnormalmetabolicprocessesandavarietyofenvironmentalfactors(i.e.,anythingthatincreasesreactiveoxygenandnitrogenspecies).Increasedlevelsof8-OH-dGareassociatedwiththeagingprocessaswellaswithanumberofpathologicalconditionsincludingcancer,diabetes,andhypertension(5-9).Incomplexsamplessuchasplasma,celllysates,andtissues,8-OH-dGcanexistaseitherthefreenucleosideorincorporatedinDNA.Oncethebloodentersthekidney,free8-OH-dGisreADIlyfilteredintotheurine,whilelargerDNAfragmentsremaininthebloodstream.Becauseofthecomplexityofplasmasamples,urineisamoresuitablematrixforthemeasurementoffree8-OH-dGthanplasma.Urinarylevelsof8-OH-dGrangebetween2.7-13ng/mgcreatine,whileplasmalevelsoffree8-OH-dGhavebeenreportedtobebetween4-21pg/mlasdeterminedbyLC-MS(10-11). |
